作者用~3H-胸腺嘧啶核苷掺入法筛选出的3株单克隆抗体(McAb)对L.d.前鞭毛体具有抑制作用,并以2B12-A8及2H6-E3作用最强,在补体协同作用下,其抑制率稳定在94~99.3%的优异水平。而1B1-C2加补体的抑制率波动于68~95%.用McAb 1B1-C2对L.d.同株前鞭毛体分别进行~3H-胸腺嘧啶核苷掺入法及巨噬细胞吞噬抑制试验,两法所得抑制率极为相近,分别为68~95%及61~87%。经免疫金技术超微定位证实,抑制作用最强的McAb 2H6-E3识别的抗原较广泛地存在于前鞭毛体膜外沿。此保护性McAb的发现,为黑热病免疫预防的研究提供了重要基础。
Three monoclonal antibodies (McAbs) screened by ~ 3H-thymidine incorporation were able to inhibit Ld promastigote and were the most potent against 2B12-A8 and 2H6-E3. Under the synergistic effect of complement , The inhibition rate stabilized at 94 ~ 99.3% of the excellent level. While the inhibition rate of 1B1-C2 plus complement fluctuated from 68% to 95%. 3Hdymithymidine incorporation and macrophage phagocytosis inhibition test were carried out on McAbs 1B1-C2 respectively. The obtained inhibition rates are very similar, respectively, 68 ~ 95% and 61 ~ 87%. Immunogold localization of the McAb 2H6-E3 confirmed that the antigens recognized by McAb 2H6-E3, which are the most potent inhibitors, are found more widely in the outer edge of the promastigote membrane. The discovery of this protective McAb provides an important basis for the study of immunoprophylaxis against kala-azar.
