黄酮类化合物能与Al3+形成稳定的荧光配合物,依此建立了同步荧光法测定三月泡叶中总黄酮的含量的新方法。用80%乙醇加热回流提取三月泡叶中总黄酮,以芦丁为标样,选择Δλ为40nm,采用导数-同步荧光光度扫描测定三月泡叶中总黄酮含量,并进行加标回收实验,验证方法的准确度和可靠性。结果表明,本方法检出限为1.27×10-9mol.L-1,建立的线性回归方程为y=0.2925x+1.1605,其相关系数r=0.9984,线性范围在0~3.3087×10-5mol.L-1之间,平均回收率0.9984。相对标准差(RSD)为0.84%。本方法操作简便、快速、准确,适应于各类可与金属离子形成荧光配合物之天然组份的快速分析。
Flavonoids can form a stable fluorescence complex with Al3+. A simultaneous fluorescence method was established to determine the content of total flavonoids in the leaves of March. The total flavonoids in foliar leaves were extracted by refluxing with 80% ethanol. The rutin was used as the standard sample. The Δλ was selected as 40 nm. The content of total flavonoids in foliar leaves of March was determined by derivative-synchronized fluorescence spectrophotometry and the recovery experiments were performed. , verify the accuracy and reliability of the method. The results show that the detection limit of this method is 1.27×10-9mol.L-1. The established linear regression equation is y=0.2925x+1.1605, and the correlation coefficient r=0.9984. The linear range is 0~3.3087×10-5mol. Between L-1, the average recovery rate was 0.9984. The relative standard deviation (RSD) was 0.84%. The method is simple, rapid, accurate and suitable for rapid analysis of various natural components that can form fluorescent complexes with metal ions.
