目的:为考察粉防己提取液致耐药大肠埃希菌外排泵表达量的下降是否由其外排泵调控基因acrR和marR基因突变引起.方法:选取临床对喹诺酮类药物耐药性较强的大肠埃希菌菌株25-1,4-5,72-1,79和质控菌ATCC25922作为受试菌,用粉防己提取液进行处理,并分析其多重耐药抑制基因marR和acrAB外排泵抑制基因acrR的突变情况.结果:耐药大肠埃希菌在粉防己提取液处理后,乳酸环丙沙星对其MIC(Minimal Inhibitory Concentration)值明显降低,且其acrR和marR基因序列较处理前有多处明显突变.结论:中药粉防己提取液可能具有逆转大肠埃希菌喹诺酮类药物耐药性的作用.
OBJECTIVE: To investigate whether the decrease of excretion pump expression of Escherichia coli-resistant Escherichia coli efflux pump is caused by the mutations of acrR and marR genes regulated by efflux pump.Methods: The drug resistance of quinolones Escherichia coli strains 25-1, 4-5, 72-1, 79 and control bacterium ATCC25922 were used as tested bacteria and treated with powder extract of Rhizoma Dioscorea and their multi-drug resistance gene marR and acrAB efflux Pump acrR gene mutation.Results: After treatment with powder extract of Escherichia coli, ciprofloxacin lactate significantly reduced its MIC (MIC), and its acrR and marR gene sequence processing There were a number of obvious mutations before .CONCLUSION: The extract of Rhizoma et Rhizoma Decoction may have the effect of reversing the resistance of Escherichia coli quinolones.
