以优良单株‘纳雍-1’的单芽茎段为外植体,建立了铁核桃(Juglans sigillata)离体培养与快速繁殖的体系。结果表明,附加6-BA 1.0 mg·L~(-1)+活性炭(AC)3.0 g·L~(-1)的DKW培养基适宜铁核桃腋芽诱导;适宜铁核桃芽增殖的培养基为DKW+6-BA1.0 mg·L~(-1)+IBA 0.02 mg·L~(-1),40 d后增殖系数可达7.33;试管苗的茎尖和茎段均可用于增殖培养;一步生根法(低浓度的生长素IBA持续诱导)不利于铁核桃试管苗嫩茎生根;采用二步生根法,生根率最高可达71.73%,其中,不同IBA浓度、暗培养时间、蔗糖浓度和AC含量对试管苗嫩茎生根影响显著,铁核桃试管苗在附加IBA 5.0 mg·L~(-1)的1/4DKW培养基中暗培养12 d,再转移到不合IBA的1/4DKW培养基(附加AC 3 g·L~(-1)和蔗糖20 g·L~(-1))中生根效果最好;生根试管苗采用珍珠岩和营养土两步炼苗,60 d后成活率达到87.50%。
In this study, a single shoot bud of excellent Nalong-1 was used as explants to establish a system of in vitro culture and rapid propagation of Juglans sigillata. The results showed that DKW medium supplemented with 6-BA 1.0 mg · L -1 + AC 3.0 g · L -1 was suitable for the induction of axillary buds of iron walnut. The suitable medium for the proliferation of iron walnut buds was DKW + 6-BA 1.0 mg · L -1 and IBA 0.02 mg · L -1, and the multiplication coefficient reached 7.33 after 40 days. The shoot tip and stem of test-tube plantlets could be used for proliferation culture. The rooting method (continuous induction with low concentration of auxin IBA) was not conducive to the rooting of the tender stem of the walnut seedlings. The rooting rate reached 71.73% by two-step rooting method. Among them, the IBA concentration, dark culture time, sucrose concentration and AC The results showed that the root of test tube had significant effects on the rooting of test-tube plantlets. The test-tube plantlets were dark-cultured for 12 days in 1 / 4DKW medium supplemented with 5.0 mg · L -1 of IBA and then transferred to 1 / 4DKW medium without IBA The rooting effect was the best with additional 3 g · L -1 of AC and 20 g · L -1 of sucrose. The rooting of test-tube seedlings was pervaded by two steps of perlite and nutrient soil. The survival rate reached 87.50 after 60 days %.
