目的研究紫外光B(UV-B)对大豆芽苗菜下胚轴中酚类物质含量的影响。方法在黑暗(D),LED(light emitting diode)白光(W)和UV-B处理下培养大豆芽苗菜。其中,W和UV-B光周期为8 h/d,光照强度为30μmol/m~2·s左右,随机取样进行下胚轴中酚类物质含量、PAL酶活及关键合成酶基因表达量的测定。结果相对D和W处理,UV-B处理显著提高了总酚物质含量和PAL酶的活性。总酚含量的提高主要体现在丁香酸、对-香豆酸和阿魏酸三种单体上。在UV-B处理下,酚类物质合成途径中四个关键酶基因PAL2、CHS8、FLS和ANS的相对表达量逐渐上升,且均在培养5 d时显著高于D和W处理。结论 UV-B通过影响大豆芽苗菜下胚轴中酚类物质合成途径相关酶基因的表达量及PAL酶活影响酚类物质含量。
Aim To study the effect of UV-B on the content of phenols in hypocotyls of soybean sprouts. Methods Soybean sprouts were cultivated under dark (D), light emitting diode (LED) and UV-B treatment. Among them, the light cycle of W and UV-B was 8 h / d and the light intensity was about 30 μmol / m ~ 2 · s. The content of phenolic compounds, the activity of PAL and the expression of key synthetase gene in hypocotyl Determination. Results Compared with D and W treatment, UV-B treatment significantly increased total phenolics content and PAL enzyme activity. The increase of total phenolic content is mainly reflected in syringic acid, p-coumaric acid and ferulic acid three monomers. Under UV-B treatment, the relative expression levels of four key enzyme genes PAL2, CHS8, FLS and ANS in the phenolic synthesis pathway were gradually increased, and were significantly higher than that of D and W treatments on the 5th day. Conclusion UV-B can affect the content of phenylalanine-lyase-related genes in the hypocotyls of soybean shoots and the PAL enzyme activity.
