目的研究导痰汤是否能够通过对内皮细胞间黏附分子-1(ICAM-1)和p38通路的调节而干预颈动脉硬化。方法新西兰白兔随机分为对照组、模型组和导痰汤大、中、小剂量组(10、8、6 g/kg导痰汤灌胃)。通过免疫组化方法,观察导痰汤对颈动脉硬化家兔血管内皮细胞ICAM-1和p38表达的影响。结果模型组血管壁内皮细胞的胞浆中有ICAM-1和p38强阳性表达,显著高于对照组(P<0.01)。导痰汤治疗组细胞内ICAM-1和p38阳性表达显著低于模型组(P<0.01),且随剂量的增加,导痰汤对ICAM-1和p38的抑制逐渐增强;而p38活性与ICAM-1水平呈显著正相关(r=0.757,P<0.01)。结论导痰汤可以有效抑制颈动脉硬化家兔血管内皮细胞中ICAM-1和p38的表达。
Objective To investigate whether Dantang Decoction can interfere with carotid atherosclerosis by regulating the expression of intercellular adhesion molecule-1 (ICAM-1) and p38. Methods New Zealand white rabbits were randomly divided into control group, model group and Dantu Tang large, medium and small dose groups (10,8,6 g / kg Guntan Tang). Immunohistochemical method was used to observe the effect of Guntan decoction on the expression of ICAM-1 and p38 in vascular endothelial cells of rabbits with carotid atherosclerosis. Results The positive expression of ICAM-1 and p38 in the cytoplasm of vascular wall endothelial cells in the model group was significantly higher than that in the control group (P <0.01). The expression of ICAM-1 and p38 in the Guntan decoction group was significantly lower than that in the untreated group (P <0.01), and with the increase of the dosage, the decoction of Yin-Tang decoction could inhibit the ICAM-1 and p38 gradually. However, -1 level was significantly positive (r = 0.757, P <0.01). Conclusion Dantan decoction can effectively inhibit the expression of ICAM-1 and p38 in the vascular endothelial cells of rabbits with carotid atherosclerosis.
