目的 研究RhoA基因mRNA在结直肠癌中的表达。方法 建立测定RhoA基因mRNA表达的逆转录聚合酶链反应 (RT PCR)最适条件 ,在半定量水平测定 42例结、直肠癌手术标本正常粘膜组织、癌组织的RhoA基因mRNA表达的相对水平 ,探讨与临床病理学指标的关系。结果 42例结、直肠癌中RhoA基因表达较相应正常粘膜组织高 ,差异有非常显著意义 (P <0 .0 1 )。以 42例肿瘤与正常粘膜表达量比值平均值为界 ,将患者分为过度表达组和高表达组 ,过度表达组中淋巴结转移发生率较高 ,肿瘤分期较晚 (P <0 .0 5)。结论 RhoA基因在结、直肠癌进展中起重要作用 ,并可能与结直肠癌浸润和转移相关
Objective To study the expression of RhoA mRNA in colorectal cancer. Methods The optimum conditions of reverse transcription polymerase chain reaction (RT PCR) for determining RhoA gene mRNA expression were established. The relative expression levels of RhoA mRNA in 42 cases of colorectal cancer, normal mucosa, To explore the relationship with clinicopathological parameters. Results The expression of RhoA gene in rectal cancer was significantly higher than that in the corresponding normal mucosa in 42 cases (P <0.01). According to the average value of 42 tumors and normal mucosa, the patients were divided into overexpression group and high expression group. The overexpression group had higher incidence of lymph node metastasis and later tumor stage (P <0.05) . Conclusion The RhoA gene plays an important role in the progression of colorectal cancer and may be related to the invasion and metastasis of colorectal cancer
