为了研究大鲵虹彩病毒(GSIV)MCP基因以及预测其B细胞表位,试验采用PCR方法扩增大鲵虹彩病毒MCP基因,采用生物学软件对大鲵虹彩病毒MCP基因的氨基酸序列进行B细胞表位预测分析。结果表明:扩增产物大小为1 392 bp,将该序列与Gen Bank中大鲵虹彩病毒MCP基因中的中国分离株核苷酸相似性为99.8%。MCP蛋白有多个抗原表位位点,抗原性指数较高,而这些区域可能是B细胞抗原表位区域。说明预测的结构、亲水性、柔性区域和抗原指数可用于大鲵虹彩病毒亚单位疫苗和分子诊断试剂的研究。
In order to study MCP gene of Rhododendron iae virus (GSIV) and to predict its B cell epitopes, PCR was used to amplify the MCP gene of Rhododendron iridiformis. The biological software was used to predict the B cell epitopes of the amino acid sequence of Rhizopus Auong ID virus MCP gene . The results showed that the size of the amplified product was 1 392 bp. The nucleotide similarity of this sequence to that of the Chinese isolate of the Actinidia virus in Gen Bank was 99.8%. MCP protein has multiple epitopes, antigenic index higher, and these areas may be B cell epitopes. It is indicated that the predicted structure, hydrophilicity, flexible region and antigen index can be used for the research on the irideski virus subunit vaccine and molecular diagnostic reagent.
