多胺生物合成抑制剂——α-二氟甲基鸟氨酸(DFMO)对人早幼粒细胞白血病细胞系HL_(60)的生长及DNA合成均有剂量依赖的抑制作用。NBT反应证明DFMO能诱导HL_(60)细胞分化。核酸分子杂交实验表明,DFMO引起c-myc基因表达的减少,而c-fos基因表达水平随DFMO浓度增加明显增加。c-myc与c-fos表达的相互变化可能与DFMO对HL_(60)细胞生长、分化影响有一定关系。
Polyamine biosynthesis inhibitor DFMO has a dose-dependent inhibitory effect on the growth and DNA synthesis of human promyelocytic leukemia cell line HL_(60). The NBT reaction demonstrated that DFMO induced differentiation of HL_(60) cells. Nucleic acid molecular hybridization experiments showed that DFMO caused a decrease in the expression of c-myc gene, and the expression level of c-fos gene increased with the increase of DFMO concentration. The mutual change of c-myc and c-fos expression may be related to the effect of DFMO on the growth and differentiation of HL_(60) cells.
