目的观察黄芩茎叶总黄酮(SSTF)对高浓度葡萄糖(HG)刺激的血管平滑肌细胞(VSMCs)增殖的抑制作用及可能的机制。方法体外培养大鼠胸主动脉VSMCs,HG诱导其增殖,观察SSTF对细胞增殖及迁移、细胞周期、醛糖还原酶(AR)mRNA及AR蛋白表达的影响。结果与模型组比较,SSTF明显抑制HG诱导的VSMCs增殖及迁移,抑制细胞周期进程(P<0.05,P<0.01),并呈时间和剂量依赖趋势;SSTF显著抑制细胞AR mRNA及AR蛋白表达(P<0.05,P<0.01)。结论 SSTF对HG刺激的VSMCs增殖具有抑制作用,可能与降低AR表达有关。
Objective To observe the inhibitory effect of SSTF on the proliferation of vascular smooth muscle cells (VSMCs) stimulated by high concentration glucose (HG) and its possible mechanism. Methods VSMCs of rat thoracic aorta were cultured in vitro. The proliferation of rat VSMCs was induced by HG. The effects of SSTF on proliferation, migration, cell cycle, aldose reductase (AR) mRNA and AR protein expression were observed. Results Compared with the model group, SSTF significantly inhibited the proliferation and migration of VSMCs induced by HG and inhibited the cell cycle progression (P <0.05, P <0.01), and showed a time-and dose-dependent trend. SSTF significantly inhibited the expression of AR mRNA and AR protein P <0.05, P <0.01). Conclusion SSTF can inhibit the proliferation of VSMCs stimulated by HG, which may be related to the decrease of AR expression.
