目的:建立一套发根农杆菌高频转化菘蓝植物的有效方法。方法:采用ATCC15834和RI1601两种发根农杆菌并通过超声波处理对草本植物菘蓝的下胚轴、子叶、带子叶下胚轴进行遗传转化,比较了共培养与否对毛状根诱导的影响,并进行了不同抗生素浓度的抑菌效果筛选和毛状根增殖培养基的筛选。结果:浸染后的外植体在不经过共培养阶段的情况下,诱导后的外植体成活率高,毛状根发根时间早,发根率高;经超声波处理后的外植体其毛状根诱导率比未处理高一倍多;同时,筛选出抗生素浓度为400 mg/L时为毛状根生长的最佳抑菌浓度;毛状根在液体培养基中的增殖速度是固体培养基的2~3倍,是普通根的37倍。结论:利用新的方法成功建立了菘蓝的毛状根诱导体系。
