目的:观察山茱萸总苷(TGCO)对大鼠实验性变态反应性脑脊髓炎(EAE)的影响。方法:Wistar大鼠24只随机分为3组:正常对照组、EAE组和TGCO组。采用动物行为学、常规苏木精-伊红染色和LFB髓鞘染色观察大鼠的发病情况与中枢神经系统(CNS)的病理变化。结果:EAE组7/8只大鼠出现典型的EAE行为学改变、CNS炎性细胞浸润和髓鞘脱失。TGCO组有3/8只大鼠出现EAE行为学改变,评分最高为2分,潜伏期延长,CNS内炎性细胞浸润和髓鞘脱失明显减轻。对照组未见动物行为学和CNS病理学改变。结论:TGCO能减轻EAE大鼠的临床症状、延长潜伏期和降低发病率。
Objective: To observe the effect of total glucosides of fructus corni (TGCO) on experimental allergic encephalomyelitis (EAE) in rats. Methods: 24 Wistar rats were randomly divided into 3 groups: normal control group, EAE group and TGCO group. Animal pathology and central nervous system (CNS) pathological changes were observed using animal behavior, routine hematoxylin-eosin staining, and LFB myelin staining. RESULTS: Typical EAE behavioral changes, CNS inflammatory cell infiltration, and myelin sheath loss were observed in 7/8 rats in the EAE group. 3/8 rats in the TGCO group had EAE behavioral changes with a score of up to 2 and prolonged latency. The inflammatory cell infiltration and myelin sheath depletion were significantly reduced in the CNS. There were no animal behaviors and CNS pathological changes in the control group. Conclusion: TGCO can reduce the clinical symptoms, prolong the incubation period and reduce the incidence of EAE rats.
