目的:选择13种巴基斯坦传统草药,观察其对PC12细胞增殖的影响,并探讨其对H2O2、谷氨酸诱导PC12细胞损伤的保护作用,为寻找神经细胞保护作用新药提供实验依据。方法:体外培养PC12细胞,应用不同浓度的H2O2、GLU,分别建立神经细胞损伤模型,采用MTT法检测细胞存活率。结果:虎爪豆、诃子等能明显促进PC12细胞增殖作用(P<0.05);在H2O2 150μmol.L-1损伤模型中,药物组和模型组相比,罗勒、仙人掌、芦笋、灯油藤、诃子等能显著减轻H2O2的细胞损伤作用(P<0.05);在GLU 30mmol.L-1诱导PC12细胞损伤,与模型组比较,凤仙花、仙人掌、虎爪豆、诃子等均显示出损伤保护作用(P<0.05)。其中,诃子不仅能促进PC12细胞增殖,且在两种PC12细胞损伤模型中均具有较强的损伤保护作用。结论:具有促进PC12细胞增殖的药物为诃子、罗勒、虎爪豆;具有对抗H2O2损伤作用的药物有:诃子、凤仙花、仙人掌、芦笋、灯油藤、罗勒;具有减弱GLU诱导PC12细胞损伤的药物为:虎爪豆、诃子、凤仙花、仙人掌、芦笋、甘草;诃子在3种模型中均表现出较强的细胞损伤保护作用。本研究结果为神经退行性疾病药物的开发和选择提供了初步的实验依据。
Objective: To select 13 kinds of Pakistani traditional herbs, observe its effect on the proliferation of PC12 cells, and to explore its protective effect on H2O2, glutamate-induced PC12 cell injury, and provide experimental basis for the search for new neuroprotective drugs. METHODS: PC12 cells were cultured in vitro. Different concentrations of H2O2 and GLU were used to establish a neuronal injury model. The cell survival rate was determined by MTT assay. RESULTS: Tilapia bean and Gardenia jasminoides could significantly promote the proliferation of PC12 cells (P<0.05). In the H2O2 150μmol.L-1 injury model, compared with the model group, basil and cactus, asparagus, and canola, Gardenia jasminoides can significantly reduce the damage of H2O2 cells (P<0.05); GLU 30mmol.L-1 induced PC12 cell damage, compared with the model group, Impatiens, cactus, tiger paw, wolfberry fruit, etc. are shown Damage protection (P < 0.05). Among them, Gardenia jasminoides not only promoted the proliferation of PC12 cells, but also had strong damage protection in both PC12 cell injury models. Conclusion: The drugs that promote the proliferation of PC12 cells are gardenia, basil, tiger paw beans; the drugs that have the effects of fighting H2O2 injury include: gardenia, impatiens, cactus, asparagus, lampau cane, basil; with reduced GLU induction of PC12 cells The drugs of injury were: Tiger Bean, Gardenia, Impatiens, Cactus, Asparagus, and Glycyrrhiza. Tillers showed strong cell damage protection in all three models. The results of this study provide a preliminary experimental basis for the development and selection of drugs for neurodegenerative diseases.
