目的探讨2型糖尿病人(T2DM)中TIM4的蛋白表达水平及与炎性小体活化指标IL-1β的相关性。方法选取新诊断T2DM患者作为病例组,正常体检者为对照,利用流式细胞术检测病人外周血单核细胞上TIM4的蛋白表达水平,同时用ELISA检测血清中可溶性TIM4的水平,同时分析TIM4与IL-1β的相关性。结果与正常对照组相比,T2DM病人血清中可溶性TIM4明显升高(P<0.001),单核细胞上TIM4表达上调(P=0.01),TIM4与IL-1β亦呈负相关性趋势(P=0.003 6,r=-0.404 5);与T2DM病人治疗前相比,治疗后外周血单核细胞上TIM4表达下降(P=0.002)。结论 T2DM病人血液中TIM4蛋白水平增高,且与IL-1β呈负相关性,提示TIM4作为负性调控分子参与T2DM的疾病进程。
Objective To investigate the expression of TIM4 in type 2 diabetes mellitus (T2DM) and its correlation with IL-1β, an inflammatory marker. Methods The newly diagnosed T2DM patients were selected as the case group and the normal subjects were taken as control. The expression of TIM4 protein in peripheral blood mononuclear cells was detected by flow cytometry. Meanwhile, the level of soluble TIM4 in serum was detected by ELISA. Meanwhile, IL-1β. Results Compared with the normal control group, the serum level of soluble TIM4 in T2DM patients was significantly increased (P <0.001), the expression of TIM4 in monocytes was up-regulated (P = 0.01), TIM4 and IL-1β also showed a negative correlation trend 0.003 6, r = -0.404 5). Compared with that before treatment, the expression of TIM4 in peripheral blood mononuclear cells decreased after treatment (P = 0.002). Conclusion The level of TIM4 in the blood of T2DM patients is increased, and it is negatively correlated with IL-1β, suggesting that TIM4 may be involved in the disease process of T2DM as a negative regulator.
