目的探讨5-氮杂-2’-脱氧胞苷(5-aza-2’-deoxycytidine,5-aza-CdR)对4种体外培养建系的黑素瘤细胞肿瘤相关抗原和免疫分子表达水平的影响。方法将手术切除或细针穿刺活组织检查获得的黑素瘤组织体外培养建系;应用流式细胞术检测5-aza-CdR用药后黑素瘤细胞表面人类白细胞抗原(human leukocyte antigen,HLA)-Ⅰ类、-Ⅱ类、-A2以及细胞间黏附分子1(intercellular adhesion molecule 1,ICAM-1)表达的变化;采用实时定量RT-PCR检测用药后一系列重要的肿瘤相关抗原表达的变化。结果 5-aza-CdR用药后,4种黑素瘤细胞系HLA-Ⅰ类、-Ⅱ类和-A2的表达与对照组相比无明显变化;TC12A和TC13细胞ICAM-1表达明显升高;TC12A、TC13和TC69B细胞肿瘤相关抗原黑素瘤抗原基因(MAGE)-4和NY-ESO-1的表达水平明显升高。结论 5-aza-CdR不影响黑素瘤细胞HLA的表达水平,对ICAM-1和某些黑素瘤相关肿瘤抗原的表达有明显上调作用,提示化疗药物5-aza-CdR不会阻碍抗肿瘤免疫中黑素瘤细胞的抗原呈递能力,为化疗联合免疫治疗肿瘤提供了理论前提。
Objective To investigate the expression of tumor associated antigens and immune molecules of melanoma cells in vitro in vitro and in vivo of 5-aza-2’-deoxycytidine (5-aza-CdR) influences. Methods Melanoma tissues obtained by surgical resection or fine needle aspiration biopsy were cultured in vitro. Flow cytometry was used to detect the expression of human leukocyte antigen (HLA) on the surface of melanoma cells after 5-aza-CdR treatment. -Ⅰ, -Ⅱ, ICAM-1 and ICAM-1 were detected by real-time quantitative RT-PCR. The expression of a series of important tumor associated antigens was detected by real-time quantitative RT-PCR. Results After 5-aza-CdR treatment, the expression of HLA-Ⅰ, -Ⅱ and -A2 in the four melanoma cell lines showed no significant changes compared with the control group; the expression of ICAM-1 in TC12A and TC13 cells was significantly increased; TC12A, TC13 and TC69B cell tumor associated antigen melanoma antigen gene (MAGE) -4 and NY-ESO-1 expression levels were significantly increased. Conclusions 5-aza-CdR does not affect the expression of HLA in melanoma cells, and significantly up-regulates the expression of ICAM-1 and certain melanoma-related tumor antigens. It suggests that 5-aza-CdR does not hinder the anti-tumor Immunization of melanoma cells antigen presenting capacity for the combination of chemotherapy and immunotherapy provides a theoretical premise of the tumor.
