目的:建立RP-HPLC测定家蚕中1-脱氧野尻霉素(DNJ)的含量。方法:采用Hypersil NH2色谱柱(4.6 mm×250mm,5μm),以乙腈-水(78∶22)为流动相,流速1.0 mL·min-1,柱温30℃,检测波长203 nm。结果:家蚕中DNJ在1.996~9.98μg(r=0.999 6)峰面积与进样量呈良好的线性关系,平均加样回收率为98.4%。结论:该方法前处理简单、实用,结果准确,可用于家蚕及制品中DNJ的质量控制。
Objective: To establish a RP-HPLC determination of silkworm 1-deoxynojirimycin (DNJ) content. METHODS: Hypersil NH2 column (4.6 mm × 250 mm, 5 μm) was used with acetonitrile-water (78:22) as the mobile phase at a flow rate of 1.0 mL · min-1 and the column temperature was set at 30 ℃ with a detection wavelength of 203 nm. Results: There was a good linear relationship between the peak area of DNJ in the range of 1.996 ~ 9.98μg (r = 0.999 6) and injection volume, with an average recovery of 98.4%. Conclusion: This method is simple, practical and accurate. It can be used for the quality control of DNJ in silkworm and its products.
