目的和方法利用BDNF、TrkB的抗体及地高辛标记的BDNFDNA探针,应用免疫组化及原位杂交方法,对正常成年大鼠腰段脊髓BDNF及其TrkB受体的分布与细胞定位进行了研究,结果BDNF及其mRNA在脊髓各型神经元中都有明显的表达,TrkB受体则主要分布在腹角运动神经元中,中间带及背角部分细胞亦有表达。胶质细胞中同时有BDNF及其mRNA、TrkB受体的表达。结论研究结果提示脊髓各型神经元,不仅是BDNF的效应细胞,其自身亦表达BDNF,靶源性、旁分泌、自分泌方式产生的BDNF共同参与了脊髓神经元的调节。胶质细胞不仅为神经元提供旁分泌因子,其自身亦受BDNF的调节。
Objectives and Methods BDNF, TrkB antibodies and digoxin-labeled BDNF DNA probes were used to detect the distribution and location of BDNF and TrkB receptors in lumbar spinal cord of normal adult rats by immunohistochemistry and in situ hybridization Results BDNF and its mRNA were all expressed in various types of spinal cord neurons. TrkB receptors were mainly distributed in the motoneuron of the ventral horn, and some cells in the medial and dorsal horn were also expressed. Glial cells in both BDNF and its mRNA, TrkB receptor expression. Conclusions The results suggest that neurons in the spinal cord are not only effector cells of BDNF, but also express BDNF, target-derived, paracrine and autocrine BDNF, which are involved in the regulation of spinal cord neurons. Glial cells not only provide paracrine factors for neurons, but are themselves regulated by BDNF.
