目的探讨不同溶剂对青龙衣中胡桃醌提取率的影响,进一步通过正交试验优选青龙衣中胡桃醌最佳提取工艺。方法以胡桃醌的量为评价指标,以乙醇、氯仿、石油醚、醋酸乙酯为溶剂分别进行超声提取,用HPLC法测定提取物中的胡桃醌,确定出最佳提取溶剂。以溶剂用量(倍)、提取时间(min)、提取次数(次)为考察因素,采用正交试验法L9(34)确定提取青龙衣中胡桃醌的最佳工艺。结果最佳提取溶剂为醋酸乙酯,最佳提取工艺条件为A3B3C2,即10倍量的醋酸乙酯,超声2次,每次40min。结论用正交试验法优选所得胡桃醌的提取工艺科学合理,适于工业生产。
Objective To investigate the effects of different solvents on the extraction rate of juglone in Qinglongyi, and to optimize the extraction of juglone from Qinglongyi by orthogonal test. Methods The amount of juglone was used as the evaluation index, and ethanol, chloroform, petroleum ether and ethyl acetate were respectively used for ultrasonic extraction. The juglone in the extract was determined by HPLC, and the optimum extraction solvent was determined. The optimum conditions of extraction of juglone in Qinglongyi were determined by orthogonal test L9 (34) with solvent amount (times), extraction time (min) and extraction times (times) as the investigation factors. Results The best extraction solvent was ethyl acetate. The optimum extraction conditions were A3B3C2, that is, 10-fold amount of ethyl acetate and sonicated twice for 40 minutes. Conclusion The optimum extraction process of jute peperone by orthogonal test is scientific and reasonable and suitable for industrial production.
