目的:探讨趋化因子及其受体CXCL16/CXCR6在人前列腺癌细胞系体外增殖侵袭中的作用。方法:免疫细胞化学技术检测CXCL16/CXCR6在人前列腺癌细胞系DU145中的表达;MTT法分析CXCL16/CXCR6对DU145细胞的促增殖作用;迁移、侵袭实验分析CXCL16对DU145细胞体外侵袭能力的调节作用。结果:DU145既表达CX-CR6蛋白,也表达CXCL16蛋白;外源性CXCL16可明显提高DU145细胞体外增殖活力和促进DU145细胞的体外迁移、侵袭能力,CXCL16中和抗体可以有效消除CXCL16对细胞的促侵袭作用,但CXCL12或CXCR4中和抗体不能阻断CXCL16的促侵袭作用。结论:CXCL16/CXCR6可能是参与前列腺癌转移的重要趋化因子配受体对。
Objective: To investigate the role of chemokine CXCL16 / CXCR6 in proliferation and invasion of human prostate cancer cell lines in vitro. METHODS: The expression of CXCL16 / CXCR6 in human prostate cancer cell line DU145 was detected by immunocytochemistry. The proliferative effect of CXCL16 / CXCR6 on DU145 cells was analyzed by MTT assay. The migration and invasion assay were used to analyze the regulatory effect of CXCL16 on the invasiveness of DU145 cells in vitro . Results: DU145 expressed CXCR6 protein and CXCL16 protein. Exogenous CXCL16 could significantly enhance the proliferation activity of DU145 cells in vitro and promote the migration and invasion of DU145 cells in vitro. CXCL16 neutralization antibody can effectively eliminate the effect of CXCL16 on cells However, CXCL12 or CXCR4 neutralizing antibodies could not block the pro-invasive effect of CXCL16. Conclusion: CXCL16 / CXCR6 may be an important chemokine receptor pair involved in the metastasis of prostate cancer.
