[目的]探讨T淋巴细胞在缺血小鼠肝脏再灌注早期浸润表达的情况。[方法]雄性C57/B6J小鼠在行肝脏部分缺血后,于再灌注15min,30min,1h,2h,4h分别处死收集肝脏标本,每个时间点7只,假手术组在相同时间点处死,每个时间点5只。用HE染色观察中性粒细胞浸润,免疫组化检测CD4+,CD8+T细胞表达。[结果]中性粒细胞在再灌注2h时才出现明显浸润,而CD4+T细胞在再灌注30min就显著高于假手术组(P﹤0.05),同时CD4+T细胞的浸润于再灌注1h到2h达到高峰。在再灌注早期各时间点,CD8+T淋巴细胞在缺血区域未发现明显浸润(与假手术组相比P﹥0.05)。[结论]在肝脏缺血再灌注早期,CD4+T淋巴细胞先于中性粒细胞进入缺血区域,并可能对中性粒细胞浸润产生调节作用。CD8+T淋巴细胞在此过程中,未起到明显作用。
[Objective] To investigate the expression of T lymphocytes in the early stage of reperfusion after liver ischemia in mice. [Methods] Male C57 / B6J mice were subjected to partial ischemia of the liver and were sacrificed at 15min, 30min, 1h, 2h, 4h after reperfusion, respectively, and liver samples were collected from each group. Seven sham operation groups were sacrificed at the same time point , Each time point 5. Neutrophil infiltration was observed by HE staining and the expression of CD4 + and CD8 + T cells was detected by immunohistochemistry. [Results] The neutrophils showed obvious infiltration at 2h after reperfusion, while the percentage of CD4 + T cells in reperfusion 30min was significantly higher than that in sham operation group (P <0.05). Meanwhile, the infiltration of CD4 + T cells in reperfusion for 1h To 2h to reach the peak. At each time point of early reperfusion, no significant infiltration of CD8 + T lymphocytes was observed in the ischemic area (P> 0.05). [Conclusion] In the early stage of liver ischemia-reperfusion, CD4 + T lymphocytes enter the ischemic area before neutrophils and may play a regulatory role in neutrophil infiltration. CD8 + T lymphocytes in the process, did not play a significant role.
