目的探讨虫草素对不同转移潜能乳腺癌细胞株TMSG-1蛋白表达的影响和TMSG-1蛋白在乳腺浸润性导管癌组织表达的意义。方法免疫组化技术检测经不同浓度的虫草素溶液(0、1、2、4μg/m)l处理24 h后,人不同转移潜能乳腺癌MCF-7、MDA-MB-231细胞爬片中TMSG-1蛋白的表达,同时检测66例乳腺浸润性导管癌组织中TMSG-1蛋白的表达,并选取癌旁组织和乳腺正常组织作为对照。结果细胞免疫组化显示,高转移潜能MDA-MB-231细胞阴性,低转移潜能MCF-7细胞阳性,虫草素2μg/ml可以上调MCF-7、MDA-MB-231细胞TMSG-1蛋白的表达,而MCF-7蛋白表达明显高于MDA-MB-231蛋白的表达,差异有统计学意义(P<0.01);66例乳腺浸润性导管癌组织中TMSG-1蛋白表达与肿瘤的淋巴结转移有明显负相关性(P<0.005)。结论虫草素2μg/ml均可以上调两个细胞株的TMSG-1蛋白表达,发挥潜在的抗肿瘤细胞浸润转移的作用。
Objective To investigate the effect of cordycepin on the expression of TMSG-1 protein in different metastatic potential breast cancer cell lines and the significance of TMSG-1 protein expression in invasive ductal carcinoma of breast. Methods Immunohistochemistry was used to detect TMSG in MCF-7 and MDA-MB-231 cells with different metastatic potentials after being treated with various concentrations of cordycepin solution (0, 1, 2, 4μg / 1 protein in 66 cases of invasive ductal breast cancer tissue TMSG-1 protein expression, and normal tissue adjacent to the breast as a control. Results Cellular immunohistochemistry showed that MCF-7 cells with high and low metastatic potential MDA-MB-231 cells were positive and cordycepin 2μg / ml could up-regulate the expression of TMSG-1 protein in MCF-7 and MDA-MB-231 cells , While the expression of MCF-7 protein was significantly higher than that of MDA-MB-231 protein, the difference was statistically significant (P <0.01). The expression of TMSG-1 protein in 66 cases of invasive ductal carcinoma of the breast with lymph node metastasis Significant negative correlation (P <0.005). Conclusion Cordycepin 2μg / ml can up-regulate the expression of TMSG-1 protein in both cell lines and play a potential role in anti-tumor cell invasion and metastasis.
