目的观察麋鹿角乙醇提取液(EAEF)对D-半乳糖(D-gal)诱导衰老模型小鼠认知功能衰退的影响。方法ICR小鼠60只,随机分成4组:正常对照组(NG),模型对照组(MG),EAEF低剂量组(LG,2g·kg-1·d-1)及高剂量组(HG,4g·kg-1·d-1)。使用D-gal按120mg·kg-1给小鼠背部sc连续6wk,造成亚急性衰老模型。造模同时,LG和HG分别ig相应浓度的EAEF,于wk7用Y型电迷宫检测小鼠学习、记忆功能,并测定小鼠脑中脂褐质(LP)含量、单胺氧化酶-B(MAO-B)活性。结果与NG比较,MG认知功能明显下降,表现为正确反应潜伏期延长,正确反应率降低(P<0.05),而EAEF能明显缩短衰老小鼠潜伏期,减少连续3次正确所需次数,显著升高正确反应率(P<0.05或P<0.01)。生化检测指标显示,MG脑组织中LP含量升高(P<0.05);EAEF能降低脑组织中MAO-B活性以及脑组织中LP含量(P<0.05)。结论EAEF对D-gal诱导衰老模型小鼠的认知功能衰退有改善作用。
Objective To observe the effect of elk ethanol extract (EAEF) on the decline of cognitive function in D-galactose-induced aging mice. Methods Sixty ICR mice were randomly divided into four groups: normal control group (MG), model control group (MG), EAEF low dose group (LG, 2g · kg -1 · d -1) and high dose group (HG, 4g · kg-1 · d-1). Mice were dosed with D-gal continuously for 6 weeks at 120 mg · kg -1, resulting in a sub-acute aging model. At the same time, LG and HG respectively ig EAEF corresponding concentration, in wk7 using Y electric maze test mice learning and memory function, and determination of mouse brain lipofuscin (LP) content, monoamine oxidase-B (MAO-B )active. Results Compared with NG, cognitive function of MG was significantly decreased, showing the correct response latency prolonged and the correct response rate decreased (P <0.05), EAEF can significantly reduce the latency of aging mice, reduce the number of consecutive 3 times the correct required, significantly increased High correct response rate (P <0.05 or P <0.01). Biochemical tests showed that LP content in MG brain tissue increased (P <0.05); EAEF decreased MAO-B activity in brain tissue and LP content in brain tissue (P <0.05). Conclusion EAEF can improve the cognitive decline of D-gal-induced aging mice.
