利用塑料细管法对9例有生育力供精者的精液进行冷冻保存,复温后在透射电镜下观察,发现精子的超微结构发生程度不同的损伤性变化。表现为精子外膜变薄、扭曲变形或破损,顶体肿胀,线粒体透亮度增加,嵴内间隙增宽,但顶体内膜和顶体中纬段仍保持完整。本文指出:冷冻复温导致精子外膜和顶体膜的通透性增加或破损,引起精子内容物漏失和线粒体功能障碍,精子能量代谢降低或停止,因而精子的活动率和运动速度在复温后低于新鲜精子。但冷冻复温并不影响精子对宫颈粘液和卵子透明带的穿透能力。
Semen of 9 fertile spermatozoa were cryopreserved by plastic tube method. After rewarming, the cells were observed under transmission electron microscope. The ultrastructural changes of spermatozoa were observed with different degrees of damage. The performance of the sperm outer thinning, distortion or damage distorted, acrosomal swelling, mitochondrial translucent increased, crest widened, but the amentoplasty and acrosome remains intact. This paper points out that the rewarming temperature increases the permeability of the spermatozoa and the acrosome membrane, resulting in the loss of sperm content and mitochondrial dysfunction, and the reduction or cessation of sperm energy metabolism. Therefore, the sperm motility and the rate of motility are increased after rewarming After less than fresh sperm. However, cryo-rewarming did not affect sperm penetration into the zona pellucida of cervical mucus and ovum.
