目的 观察重组人睫状神经营养因子(rhCNTF)对大鼠胶质瘤C6细胞系体外生长的影响。方法 MTT、软琼脂克隆试验和流式细胞仪检测C6细胞体外增殖、克隆形成和细胞周期。结果 20~2000 ng/ml rhCNTF可明显抑制C6细胞的增殖,其克隆形成率降低显著,G_1、G_2期细胞明显增多,S期细胞显著减少。结论 rhCNTF对C6细胞的生长抑制作用可能与其参与瘤细胞的生长调节有关,表明CNTF作为一种细胞调节因子对胶质瘤具有潜在治疗作用。
Objective To observe the effect of recombinant human ciliary neurotrophic factor (rhCNTF) on the growth of rat glioma C6 cell line in vitro. Methods MTT, soft agar cloning and flow cytometry were used to detect the proliferation, colony formation and cell cycle of C6 cells in vitro. Results The proliferation of C6 cells was inhibited by 20-2000 ng/ml rhCNTF, and the colony formation rate was significantly decreased. The cells in G 1 and G 2 phases were significantly increased, and the cells in S phase were significantly decreased. Conclusion The inhibitory effect of rhCNTF on the growth of C6 cells may be related to its involvement in the growth regulation of tumor cells, indicating that CNTF as a cell regulator has a potential therapeutic effect on gliomas.
