目的研究核糖核酸酶抑制因子对小鼠实体瘤生长的影响以及RI抑制肿瘤生长的机理。方法经硫酸铵盐析,DEAE-离子交换层析和Sepharose-RNase亲和层析,从人胎盘中提取纯化核糖核酸酶抑制因子(ri-bonucleaseinhibitor,RI),将其注射给荷瘤小鼠。5天后处死小鼠,取瘤组织,称重;组织切片后观察瘤内毛细血管、组织坏死状况及核分裂数的改变。结果注射RI后,小鼠实体瘤S180的生长受到抑制,瘤组织毛细血管减少,坏死加强。结论RI抑制肿瘤的血管生成,从而抑制实体瘤的生长
Objective To investigate the effect of ribonuclease inhibitors on the growth of solid tumors in mice and the mechanism of inhibition of tumor growth by RI. Methods The ribonuclease inhibitor (RI) was extracted from human placenta by ammonium sulfate precipitation, DEAE-ion exchange chromatography and Sepharose-RNase affinity chromatography, and injected into tumor-bearing mice. Five days later, the mice were sacrificed, and the tumor tissues were removed and weighed. After the tissue sections were taken, the changes of the capillaries, tissue necrosis, and the number of mitotic cells were observed. Results After the injection of RI, the growth of S180 solid tumor was inhibited, and the capillaries in the tumor tissue decreased and the necrosis enhanced. Conclusion RI inhibits angiogenesis of tumors and inhibits the growth of solid tumors
