建立香草醛 磷酸体系荧光分光光度法测定黄芪甲甙的方法。方法 :利用在磷酸条件下香草醛与黄芪甲甙的荧光反应测定黄芪甲甙。结果 :黄芪甲甙与香草醛的反应产物的最大激发波长Ex =36 5nm ,最大发射波长Em =430nm。黄芪甲甙在 0 .0~ 2 0 .0mg·L-1范围呈良好的线性关系 ,最低检测限为 0 .10 6mg·L-1,测定样品的回收率为 95 .97%~ 10 2 .1%。结论 :本方法具有灵敏度高、检测限低、直接测定无干扰等优点。测定黄芪口服液、中药复方补阳还五汤及含药猪血清等多种类型样品中的黄芪甲甙含量 ,结果满意。
A method for the determination of Astragaloside A by fluorescence spectrophotometry with vanillin phosphate system was established. Methods: The fluorescence reaction of vanillin and Astragaloside in the presence of phosphoric acid was used to determine Astragaloside. RESULTS: The maximum excitation wavelength of the reaction product of Astragaloside and vanillin was Ex = 36 5 nm, and the maximum emission wavelength was Em = 430 nm. Astragaloside IV showed a good linear relationship in the range of 0.0 to 20.0 mg·L-1, with a detection limit of 0.16 mg·L-1. The recovery of the assay sample was 95.97% to 10 2 . 1%. Conclusion : This method has the advantages of high sensitivity, low detection limit and direct measurement without interference. The results showed that the content of Astragaloside in various types of samples such as Huangqi Oral Liquid, Chinese Herbal Compound Buyang Huanwu Decoction, and serum containing drug-containing pigs was satisfactory.
