目的:探讨扶正化瘀方抗肾间质纤维化的主效药物及配伍特点。方法:除正常对照组外,其余大鼠采用氯化汞(HgCl2)灌胃诱导肾间质纤维化模型。采用正交实验设计,根据L16(215)正交表设为16个组,其中给药者15组,分别按正交表药物组合给药;无药物者为模型组。药物干预后,分别观察各组大鼠体重、肾重,肾组织炎症与胶原沉积变化,检测肾组织羟脯氨酸(Hyp)含量,血清尿素氮(BUN)、肌酐(Scr)及尿蛋白浓度。结果:升高大鼠体重起主效应的药物是虫草菌丝、桃仁,有明显交互作用的是丹参/桃仁;降低肾体重比起主效应的药物是丹参、松黄、绞股蓝,有明显交互作用的是虫草菌丝/五味子;降低纤维化肾组织Hyp含量起主效应的药物是丹参、松黄、虫草菌丝、桃仁,有明显交互作用的是虫草菌丝/五味子、丹参/五味子、虫草菌丝/丹参、绞股蓝/五味子;降低BUN起主效应的是丹参、虫草菌丝、松黄、桃仁,有明显交互作用的是虫草菌丝/五味子、虫草菌丝/丹参;降低Scr起主效应的是虫草菌丝、松黄,呈明显交互作用的有丹参/桃仁;降低尿蛋白浓度的主效应药物是丹参、虫草菌丝、五味子,且考虑的6个交互作用都有统计意义。结论:扶正化瘀方抗大鼠肾纤维化的主要有效药物为丹参、虫草菌丝、松黄、桃仁;尤其以丹参作用突出,为全方之君药,与虫草菌丝、桃仁有良好交互作用,因此活血化瘀与益精补虚药物配伍具有综合疗效优势。
Objective: To investigate the main effects of Fuzheng Huayu recipe anti-renal interstitial fibrosis and its compatibility characteristics. Methods: In addition to the normal control group, other rats were induced by intragastric administration of mercury chloride (HgCl2) into the model of renal interstitial fibrosis. Orthogonal experimental design was adopted. According to the L16 (215) orthogonal table, 16 groups were set up, of which 15 were administered group, which were administered by orthogonal test. Drug-free group was model group. The changes of body weight, kidney weight, inflammation of renal tissue and collagen deposition in rats were observed after drug intervention. The contents of hydroxyproline (Hyp), serum urea nitrogen (BUN), creatinine (Scr) . Results: The drug that lifted the body weight of rats was Cordyceps sinensis mycelia and peach kernel, and there was significant interaction between Salvia miltiorrhiza and Peach kernel. The drug with lower body weight than the main effect was Salvia miltiorrhiza, Is Cordyceps mycelium / Schisandra; reduce Hyp fibrosis renal tissue main effect of the drug is Salvia miltiorrhiza yellow, Cordyceps mycelium, peach kernel, there is a clear interaction between Cordyceps / Schisandra, Salvia / Schisandra, Cordyceps mycelium / Salvia miltiorrhiza, Gynostemma pentaphylla / Schisandra; the main effect of reducing the main effect of BUN is Salvia miltiorrhiza, Cordyceps mycelium, pine yellow, peach kernel, obvious interaction is Cordyceps mycelium / Schisandra, Cordyceps mycelium / Salvia; Cordyceps sinensis, pine yellow, showed significant interaction with Salvia / peach kernel; to reduce the main effect of urinary protein concentration drugs are Salvia, Cordyceps mycelium, Schisandra, and consider the six interactions have statistical significance. Conclusion: The main effective drugs of Fuzheng Huayu Recipe for inhibiting renal fibrosis in rats are Radix Salviae Miltiorrhizae, Cordyceps mycelium, pine yellow and peach kernel, especially Salvia miltiorrhiza, which is the monarch drug of all prescriptions and has good interaction with Cordyceps mycelium and peach kernel. Therefore, promoting blood circulation and essence of tonic drug compatibility has a comprehensive efficacy advantage.
