目的研究姜辛夏颗粒对卵蛋白(OVA)致敏哮喘大鼠模型肺组织中Ⅰ、Ⅲ、Ⅴ型胶原蛋白表达及对支气管壁厚度的影响。方法采用OVA致敏建立哮喘大鼠模型,观察姜辛夏颗粒对哮喘大鼠的疗效;免疫组化技术检测肺组织中Ⅰ、Ⅲ、Ⅴ型胶原蛋白的表达;支气管组织HE染色,计算机图像分析测定气道壁厚度。结果与模型组比较,姜辛夏颗粒能明显减轻哮喘大鼠的哮喘症状和支气管黏膜的炎症反应;明显缩小哮喘大鼠支气管总管壁面积(WAt)、内壁面积(WAi)及平滑肌面积(WAm)(P<0.01);明显降低肺组织中I、Ⅲ、V型胶原蛋白的表达(P<0.01,P<0.05)。结论姜辛夏颗粒可明显降低哮喘大鼠支气管上皮下Ⅰ、Ⅲ、Ⅴ型胶原蛋白的合成和沉积,减小支气管壁的厚度,改善支气管的通气功能,从而抑制哮喘气道重建的发生发展。
Objective To investigate the effects of Jiangxinxia Granules on the expression of collagen type Ⅰ, Ⅲ and Ⅴ in bronchial asthma rat model of asthma induced by ovalbumin (OVA) and its effect on bronchial wall thickness. Methods Asthma rat model was established by OVA sensitization. The therapeutic effects of Jiangxinxia granule on asthmatic rats were observed. The expression of collagen type Ⅰ, Ⅲ and Ⅴ in lung tissue was detected by immunohistochemistry. The bronchial tissue was stained with HE and analyzed by computer image analysis Road wall thickness. Results Compared with the model group, Jiangxinxia Granules could significantly reduce the asthma symptoms and bronchial mucosal inflammatory response in asthmatic rats; significantly reduce the bronchial wall area (WAt), the internal wall area (WAi) and the smooth muscle area (WAm) P <0.01). The expression of collagen type I, III and V in lung tissue was significantly decreased (P <0.01, P <0.05). Conclusion Jiangxinxia granule can significantly reduce the synthesis and deposition of type Ⅰ, Ⅲ and type Ⅴ collagen in the bronchial epithelium of asthmatic rats, reduce the thickness of the bronchial wall and improve the bronchial ventilation function, thus inhibiting the occurrence and development of airway remodeling in asthma.
