应用PAGE从粉尘螨Dermatophagoides farinae浸液中分离出11条蛋白质区带。挑刺试验证明,其中7条区带的洗脱液含有过敏原,能使171例吸入型哮喘病人呈阳性反应,以6号区带洗脱液的过敏反应最强,阳性率达49.1%。全部区带洗脱液不能使所有病人都呈现阳性反应;每一过敏患者仅对其中某些区带的洗脱液出现过敏。用对流免疫电泳(CIE)证明3种区带洗脱液含有沉淀抗原,但只合少量过敏原,过敏性最强的6号区带洗脱液不含沉淀抗原。用不连续PAGE从粉尘螨浸液中分离出21种蛋白质组分,其中6种含糖蛋白,4种含脂蛋白。应用SDS-PAGE可测得40余种蛋白质组分。
Eleven protein bands were isolated from the extracts of Dermatophagoides farinae by PAGE. The prick test proved that seven of the strips contained allergens, which could make 171 patients with inhaled asthma positive reaction. The strongest allergen response was obtained with zone 6 eluent. The positive rate was 49.1%. Entire zone eluents do not give a positive response to all patients; each allergic patient is allergic to only some of the zones. Convergent immunoelectrophoresis (CIE) demonstrated that the three zone eluents contained precipitated antigens, but only a small amount of allergens and the most hypersensitive zone 6 eluents did not contain precipitated antigens. Twenty-one protein fractions were isolated from Dust-Dermatophagoides farinae by discontinuous PAGE, of which six were glycoproteins and four were lipoproteins. More than 40 kinds of protein components can be measured by SDS-PAGE.
