目的:探讨抑郁模型大鼠海马CA3区突触结构的体视学变化及加味温胆汤的作用。方法:以孤养结合慢性不可预见性应激抑郁模型大鼠为研究对象,在实验第7d,14d,21d,28d,检测各组大鼠海马突触结构的体视学变化。结果:在抑郁形成过程中,21d时,与空白组比较,模型组大鼠突触活性区长度、突触后致密物质厚度明显减少,突触间隙宽度增加明显。28d时,差异进一步明显。随着加味温胆汤(12g/kg)治疗周次的增加,抑郁大鼠突触活性区长度、突触后致密物质厚度减少,以及突触间隙宽度上升的趋势得到抑制,与模型组比较,在21d时,有明显统计学差异(P<0.05),28d时,差异增大(P<0.01)。加味温胆汤汤组(12g/kg)与氟西汀组(1.8mg/kg)相比无明显统计学意义。结论:抑郁症可引起海马神经元突触结构损伤,导致出现神经元萎缩,加味温胆汤对此有逆转作用。
Objective: To investigate the stereological changes of synaptic structure in hippocampal CA3 area of depression model rats and the effect of Jiawei Wendan Decoction. Methods: To investigate the stereological changes of synaptic structure in the hippocampus of rats in each group by solitary combined with chronic unpredictable stress depression model rats at 7d, 14d, 21d and 28d. Results: Compared with the blank group, the length of synaptic active area and the thickness of the postsynaptic dense substance in the model group decreased significantly and the width of the synaptic cleft increased significantly in the process of depression. 28d, the difference is further obvious. With the increase of treatment week of Jiawewewendan Decoction (12g / kg), the length of synaptic active area, the decrease of the thickness of the postsynaptic density and the increase of the width of synaptic cleft were inhibited in the depressed rats. Compared with the model group, There was a significant difference between the two groups (P <0.05) at 21 d, and at 28 d (P <0.01). Modified Wei Dan Tang Decoction group (12g / kg) and fluoxetine group (1.8mg / kg) compared with no statistically significant. Conclusion: Depression can cause synaptic structural damage in hippocampal neurons, leading to the appearance of atrophy of neurons. Jiawei Wendan Decoction reverses this effect.
