[目的]探究北京红地球葡萄不同部位内生菌的种类及多样性。[方法]对北京市葡萄主产区种植的红地球葡萄叶片、果梗、果肉、果皮各部位进行内生菌的分离、鉴定和多样性分析。[结果]红地球葡萄不同部位的内生菌数量存在差异,果梗中内生细菌数量最多,果肉中内生真菌数量最多;共分离获得内生细菌11株,16S r DNA序列鉴定结果表明果肉中内生细菌种类多样性高于果梗、果皮和叶片,芽孢杆菌属(Bacillus sp.)在各组织中均有分布;变性梯度凝胶电泳(DGGE)结果表明不同部位内生细菌菌群存在差异性。[结论]试验结果为葡萄内生微生态系统多样性的深入研究提供了理论依据。
[Objective] The research aimed to explore the species and diversity of endophytes in different parts of Beijing Red Globe grape. [Method] The isolation, identification and diversity analysis of endophyte from leaves, fruit stems, flesh and peel of Red Globe grape planted in main grape growing areas of Beijing were analyzed. [Result] The numbers of endophytic bacteria were different in different parts of Red Globe Grape. The endophytic bacteria in fruit stems were the most, and the endophytic fungi in fruit were the most. There were 11 endophytic bacteria isolated in total, and 16S rDNA sequence identification results showed that the flesh The endophytic bacteria species diversity was higher than that of fruit stems, peels and leaves, and Bacillus sp. Was distributed in all tissues. The results of denaturing gradient gel electrophoresis (DGGE) showed that endophytic bacteria colonies existed in different parts difference. [Conclusion] The results provided the theoretical basis for the further study of the diversity of endophytic micro-ecosystems in grapes.
