目的探讨酪氨酸蛋白磷酸酶样A结构域2(PTPLAD2)基因产物表达与食管鳞状细胞癌(ESCC)临床病理特征及预后的关系。方法应用免疫组织化学染色法分析了213例ESCC标本中PTPLAD2基因产物表达情况和定位,并对其进行了术后5 a随访。结果 PTPLAD2主要定位在细胞膜,且其在正常鳞状上皮中表达较高(54.0%),而在ESCC组织中低表达(16.0%),对PTPLAD2的表达与ESCC患者临床病理特征关系分析结果显示,在有淋巴结转移和预后差的ESCC病例中低表达;PTPLAD2在ESCC中的表达与ESCC的浸润、转移及分化程度均相关(P<0.05);PTPLAD2表达可作为ESCC患者预后独立指标。结论 PTPLAD2在ESCC中的表达与食管癌的发生发展、浸润转移和预后密切相关,PTPLAD2可能是ESCC发生相关的新的潜在抑癌基因,可作为临床预测浸润转移和估计预后的一个重要参考指标。
Objective To investigate the relationship between the expression of tyrosine protein phosphatase-like domain 2 (PTPLAD2) gene and the clinicopathological characteristics and prognosis of esophageal squamous cell carcinoma (ESCC). Methods The expression and localization of PTPLAD2 gene in 213 cases of ESCC were analyzed by immunohistochemical staining. The patients were followed up for 5 years. Results PTPLAD2 mainly localized in the cell membrane and its expression was higher in normal squamous epithelium (54.0%) than in ESCC tissues (16.0%). Analysis of the relationship between PTPLAD2 expression and clinicopathological features of ESCC showed that, The expression of PTPLAD2 in ESCC was correlated with the degree of invasion, metastasis and differentiation of ESCC (P <0.05). The expression of PTPLAD2 was an independent prognostic factor in patients with ESCC. Conclusion The expression of PTPLAD2 in ESCC is closely related to the carcinogenesis, invasion, metastasis and prognosis of esophageal carcinoma. PTPLAD2 may be a new potential tumor suppressor gene associated with ESCC, which may serve as an important reference for clinical prediction of invasion and metastasis and prognosis.
