目的 :建立同时测定血清中苯巴比妥、苯妥英钠和卡马西平的浓度的高效液相色谱法 (HPLC)。方法 :血清乙酸乙酯提取浓集后进样 ,采用C18柱 ( 2 2 0× 4 .6,5μm) ,以甲醇 -水 ( 50∶50 )为流动相 ,苯乙酮为内标物 ,在波长2 1 0nm处测定。结果 :苯巴比妥、苯妥英钠和卡马西平的线性范围为 0 .63~ 4 4μg/ml,三者平均回收率均大于 97% ,日内和日间误差小于 5%。结论 :本法快速、简便、准确 ,适用于治疗药物监测和药动学研究。
Objective: To establish a high performance liquid chromatography (HPLC) method for the simultaneous determination of phenobarbital, phenytoin and carbamazepine in serum. Methods: Serum ethyl acetate was extracted and concentrated before injection. C18 column (2 2 0 × 4. 6 and 5 μm), methanol-water (50:50) as mobile phase and acetophenone as internal standard Wavelength of 210nm at the determination. Results: The linear ranges of phenobarbital, phenytoin sodium and carbamazepine were 0.63-4.4μg / ml. The average recoveries of all three were more than 97%, and the error between day and day was less than 5%. Conclusion: This method is rapid, simple and accurate and is suitable for drug monitoring and pharmacokinetics research.
