建立了胸腺五肽含量的HPLC检测法,采用C18柱,流动相为50mmol/L磷酸二氢钾溶液(pH7.0)-甲醇(85∶15);流速1ml/min;检测波长275nm。考察了胸腺五肽在不同温度、pH、缓冲盐体系中的稳定性。结果表明,胸腺五肽在酸性介质中降解速度加快。磷酸缓冲体系较柠檬酸缓冲体系更有利于保持胸腺五肽的稳定。
A HPLC method was developed for the determination of thymopentin. The mobile phase consisted of 50mmol / L potassium dihydrogen phosphate (pH7.0) -methanol (85:15) with a C18 column at a flow rate of 1ml / min. The detection wavelength was 275nm. The stability of thymopentin in different temperature, pH and buffer system was investigated. The results showed that the degradation of thymopentin in acidic medium accelerated. Phosphate buffer system is more conducive to maintaining the stability of thymopentin than citrate buffer system.
