目的:该研究探讨维甲酸对Bel7402人肝癌细胞的分化诱导作用,为肝癌的分化治疗提供资料。方法:用维甲酸处理Bel7402人肝癌细胞,观察细胞增殖、软琼脂集落形成和各项生化指标的变化。结果:10μmol/L维甲酸显著抑制肝癌细胞增殖,并使肝癌细胞软琼脂集落形成率明显减少,使代表肝细胞分化的酶鸟氨酸氨基甲酰转移酶、酪氨酸α酮戊二酸转氨酶和碱性磷酸酶比活力明显升高,甲胎蛋白分泌量、γ谷氨酰转肽酶和醛缩酶比活力明显下降。结论:证明维甲酸是Bel7402人肝癌细胞有效的分化诱导剂。
OBJECTIVE: To investigate the effect of retinoic acid on the differentiation of Bel7402 human hepatocellular carcinoma cells and to provide data for the differentiation and treatment of hepatocellular carcinoma. METHODS: Bel7402 human hepatocellular carcinoma cells were treated with retinoic acid, and cell proliferation, soft agar colony formation and changes in biochemical parameters were observed. RESULTS: Retinoic acid at 10 μmol/L significantly inhibited the proliferation of hepatocellular carcinoma cells and reduced the formation rate of soft agar colonies. The results showed that the ornithine carbamoyltransferase and tyrosine α-ketoglutarate transaminase, which represent the differentiation of hepatocytes The specific activity of alkaline phosphatase was significantly increased, and the secretion of alpha-fetoprotein and the specific activity of gamma glutamyl transpeptidase and aldolase decreased significantly. Conclusion: It is proved that retinoic acid is an effective differentiation inducer for Bel7402 human hepatoma cells.
