目的研究人血浆萘哌地尔的高效液相色谱(HPLC)测定。方法血浆样品依次用6mol/LHCl、NaOH处理,乙醚提取2次。色谱分析柱为粒径5μm填料为EcosphereC18,内标为美托洛尔,流动相为甲醇-乙腈-水-0.02mol/LKH2PO4(55∶20∶22∶3),pH4.10。荧光检测器激发波长290nm,发射波长340nm。结果最低检测浓度为1.0ng/ml,天内及天间的变异系数为1.15%至4.71%。结论此法价廉、灵敏、可靠性及重复性好,故适合国内条件下研究人体药代动力学。
Objective To study the determination of naftopidil in human plasma by high performance liquid chromatography (HPLC). Methods Plasma samples were treated with 6mol / LHCl, NaOH and extracted twice with ether. The chromatographic column was Ecosphere C18 with a particle size of 5 μm and metoprolol internal standard. The mobile phase was methanol-acetonitrile-water-0.02 mol / L KH 2 PO 4 (55:20:22:3), pH 4.10. Fluorescence detector excitation wavelength 290nm, emission wavelength 340nm. Results The minimum detectable concentration was 1.0 ng / ml. The coefficient of variation of intra-and inter-day was 1.15% -4.71%. Conclusion This method is cheap, sensitive, reliable and reproducible, so it is suitable for the study of human pharmacokinetics under domestic conditions.
