建立MTT(3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazoliumbromide)比色法进行血小板免疫活性的测定。最适血小板浓度为107/孔,最适MTT浓度为4mg/ml(80μg/孔)。IgE依赖组与阿斯匹林依赖组血小板活性测定MTT比色法与以往的血小板介导的杀寄生虫幼虫的细胞毒素测定方法呈显著相关(IgE依赖组两者的相关系数r=0951,阿斯匹林依赖组两者的相关系数r=0802)。实验表明,MTT比色法因其方法简便、准确、试剂易得,可作为临床检测血小板活性的一种实用方法
The establishment of MTT (3- (4,5-dimethylthiazol-2-yl) -2,5-diphenyl-tetrazoliumbromide) assay for platelet immunocompetence. The optimal platelet concentration was 107 / well, and the optimal MTT concentration was 4mg / ml (80μg / well). MTT colorimetric assay was significantly associated with platelet-mediated killing of parasitic larvae in the IgE-dependent and aspirin-dependent groups (IgE-dependent group, r = 0.951 , Aspirin dependence group correlation coefficient r = 0802).
