目的 :探讨不同浓度益生注射液A、B两种组分及联合用药对移植物慢性失功 (CAD)的防治效果及作用环节。方法 :利用组织形态观察 ;计算机图象分析 ;过氧化脂质测定和免疫组织化学分析大鼠腹主动脉移植硬化加快模型。结果 :术后 6周A +B及骁悉 (MMF)治疗组能显著抑制血管内膜的增厚 ,B( 10mg .kg 1)、B( 1mg .kg 1)、A+B组术后各期过氧化脂质 (LPO)与术前比无明显差异 ,各组TGF β1表达高峰在术后 2周 ,空白对照组各时间点的表达最强 ,A +B及MMF组表达最弱。结论 :A组分通过抑制T、B淋巴细胞及巨噬细胞增殖、浸润 ,间接下调TGF β1的表达 ,B组分对氧自由基引起的损伤有明显保护作用 ,A +B有协同作用 ,其作用与MMF单用相当
Objective : To investigate the effect of different concentrations of probiotics A and B components and combination therapy on the prevention and treatment of chronic graft failure (CAD). Methods : Using tissue morphology observation; computer image analysis; lipid peroxide assay and immunohistochemical analysis of rat abdominal aortic graft hardening model. RESULTS: At 6 weeks postoperatively, the A + B and MMF treatment groups significantly inhibited the intimal thickening of the blood vessels. B (10 mg.kg 1), B (1 mg.kg 1), and A+B groups were postoperative. There was no significant difference in lipid peroxidation (LPO) between preoperative and postoperative period. The peak expression of TGFβ1 in each group was at 2 weeks after operation. The expression of TGFβ1 in the blank control group was the strongest at all time points, and that of A+B and MMF groups was the weakest. Conclusion: A-component can indirectly down-regulate the expression of TGFβ1 by inhibiting the proliferation and infiltration of T and B lymphocytes and macrophages. The B component has obvious protective effect on the damage caused by oxygen free radicals. A + B has a synergistic effect. The effect is equivalent to MMF alone
