姜黄素预处理对大鼠肾脏缺血再灌注损伤的影响及其机制

目的:探讨姜黄素对大鼠肾脏缺血再灌注损伤(IRI)细胞因子的影响。方法:制备大鼠肾IRI模型,双抗体夹心ELISA法动态测定肾组织和血液中肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)含量,并检测尿素氮(BUN)、肌酐(Cr)指标变化,计算肾系数。结果:单纯肾缺血再灌注1 h(R1h)组,肾组织中TNF-α水平明显增高,随再灌注时间延长逐渐下降至假手术对照组水平;血清TNF-α在IRI各组均无显著性变化。血液和肾组织中IL-6含量在单纯缺血组明显降低(P<0.05),而R1h组又升至对照组水平。缺血再灌注4 h(R4h组)和缺血再灌注24h组(R24h组)肾组织中的IL-6明显低于R1h组和对照组(P<0.05);IL-6含量的动态变化在肾组织和血液中呈直线正相关(r=0.846,P<0.01)。姜黄素干预,可使R4h和R24h组肾组织中的IL-6含量明显增高;同时使R4h组血清BUN、Cr及肾系数也明显高于未干预组,而R24h组的Cr明显降低。姜黄素干预后,血清和肾组织中TNF-α无显著性变化。结论:TNF-α、IL-6在早期大鼠肾IRI中起重要作用,姜黄素预处理早期可使IL-6含量升高,加重肾功能损害,随着再灌注时间延长,肾功能逐渐好转。 Objective: To investigate the effect of curcumin on cytokines in renal ischemia-reperfusion injury (IRI) in rats. Methods: IRI model of rat kidney was prepared. The contents of TNF-α and IL-6 in renal tissue and blood were measured by double antibody sandwich ELISA. The levels of blood urea nitrogen (BUN) ), Creatinine (Cr) index changes, calculate the renal coefficient. Results: The levels of TNF-α in renal tissue were significantly increased at 1 h (R 1h) and decreased gradually to the sham-operated control group with prolonged reperfusion time. Serum TNF-α levels were not significantly different in IRI groups Sexual change. The levels of IL-6 in blood and kidney tissue were significantly decreased in the ischemia group (P <0.05), while those in the R1h group increased to the control group. The levels of IL-6 in renal tissue in ischemia reperfusion 4 h (R4h group) and ischemia reperfusion 24h group (R24h group) were significantly lower than those in R1h group and control group (P <0.05). The dynamic changes of IL- There was a linear positive correlation between renal tissue and blood (r = 0.846, P <0.01). Curcumin intervention could significantly increase the levels of IL-6 in renal tissues of R4h and R24h groups, meanwhile, the levels of BUN, Cr and renal index in R4h group were significantly higher than those in non-intervention group, while Cr in R24h group was significantly lower. After the intervention of curcumin, there was no significant change of TNF-α in serum and kidney tissue. CONCLUSIONS: TNF-α and IL-6 play an important role in early renal IRI. Curcumin pretreatment can increase the content of IL-6 in early stage and aggravate renal dysfunction. With the reperfusion time prolonging, renal function gradually improves .
