目的探讨甲状腺正常组织和分化型甲状腺癌(DTC)中β-catenin和Cyclin D1的表达及临床意义。方法应用免疫组织化学链霉菌抗生素蛋白-过氧化物酶连接法(SP法)检测10 例腺瘤旁正常甲状腺组织和57例DTC标本中β-catenin和Cyclin D1的表达。结果β-catenin在甲状腺正常组织中均为正常表达,Cyelin D1均呈阴性。DTC中β-catenin和Cyclin D1的异常表达率分别为57.9%和64.9%。DTC中β-catenin异常表达与淋巴结转移密切相关;Cyclin D1表达与淋巴结转移和病理高分期有关。DTC中β-catenin和Cyclin D1表达呈明显正相关(P<0.01)。结论β-catenin异常表达可能激活靶基因Cyclin D1的表达,并在甲状腺癌发生发展中起到重要作用。
Objective To investigate the expression and clinical significance of β-catenin and Cyclin D1 in normal thyroid tissue and differentiated thyroid carcinoma (DTC). Methods The expressions of β-catenin and Cyclin D1 in 10 normal thyroid tissues and 57 DTC specimens were detected by SP immunohistochemical streptavidin-peroxidase method (SP method). Results β-catenin in normal thyroid tissue were normal expression, Cyelin D1 were negative. The abnormal expression rates of β-catenin and Cyclin D1 in DTC were 57.9% and 64.9% respectively. The abnormal expression of β-catenin in DTC is closely related to the lymph node metastasis. The expression of Cyclin D1 is related to lymph node metastasis and high pathological stage. The expression of β-catenin and Cyclin D1 in DTC was positively correlated (P <0.01). Conclusion The abnormal expression of β-catenin may activate the expression of the target gene Cyclin D1 and play an important role in the occurrence and development of thyroid cancer.
