目的 :用GC/MS/MS法测定尿中 11-羟基 -△9-四氢大麻酚二酸 (THC -COOH)的浓度。滥用大麻后 ,主要起精神兴奋作用的是 11-羟基 -△9-四氢大麻酚二酸 (THC -COOH) ,它也是鉴定滥用大麻的依据。GC/MS/MS提供了灵敏可靠的鉴定技术。方法 :本方法在进入GC/MS/MS分析之前 ,尿样要进行碱水解 (10 %KOH甲醇溶液 ) ,以破坏其结合型 ,然后用浓盐酸HCl和冰醋酸HAc调至pH =3.5左右 ,加 2ml正庚烷提取 3次 ,在N2 下吹干。残渣衍生化后 ,进 1μl于GC/MS/MS进行分析。结果 :本方法能灵敏地检测到 15ng/ml左右的THC -COOH。结论 :满足IOC检测要求
OBJECTIVE: To determine the concentration of 11-hydroxy-? 9-tetrahydrocannabenol diacid (THC-COOH) in urine by GC / MS / MS. After abuse of marijuana, 11-hydroxy-? 9-tetrahydrocannabenol (THC-COOH), which is primarily a psychoactive substance, is also the basis for identification of abuse of marijuana. GC / MS / MS provides sensitive and reliable identification techniques. Methods: Prior to GC / MS / MS analysis, urine samples were subjected to alkaline hydrolysis (10% KOH in methanol) to disrupt their binding and then adjusted to pH = 3.5 using concentrated HCl and HAc. Add 2ml n-heptane extract 3 times, dry under N2. After the residue was derivatized, 1 μl was analyzed by GC / MS / MS. Results: The method can sensitively detect about 15ng / ml THC-COOH. Conclusion: Meet IOC testing requirements
