目的研究冷、热两种工艺加工的鹿茸提取物,对正常、慢性再生障碍性贫血患者骨髓的单个核细胞(mononuclear cell,MNC)体外增殖、分化的影响,为临床合理应用提供实验依据。方法从10例健康者、10例慢性再生障碍性贫血(aplastic anemia,AA)患者骨髓中获取MNC,分别在普通培养液(阴性对照组),加热炸茸C组分的培养液(试验组1)及加冷工艺茸A、B、D、E组分的培养液(试验组2)中培养。MTT比色法检测细胞的增殖活性;CFU-mix集落半固体培养观察集落生长情况;细胞免疫化学染色测定bcl-2、突变型p53的表达。数据处理采用单因素方差分析(one-way ANOVA),结果用x珋±s表示。结果正常骨髓组:试验组2细胞增殖活性及CFU-mix集落数均明显高于试验组1及阴性对照组;bcl-2和突变型p53的表达各组之间均无明显差异。AA骨髓组:试验组2中D、E组分细胞增殖活性和CFU-mix集落数均明显高于阴性对照组及试验组1。Bcl-2的表达:试验组2中D、E组分组显著高于阴性对照组和试验组1。突变型p53的表达三组之间均无明显差异。结论两种不同工艺的鹿茸相比,冷工艺茸具有更强的生物学效应,故在鹿茸的加工上推荐冷工艺。
Objective To study the effects of cold and hot process antler extracts on the proliferation and differentiation of mononuclear cells (MNC) in the bone marrow of patients with normal and chronic aplastic anemia, and to provide experimental basis for its clinical application. Methods MNCs were obtained from bone marrow of 10 healthy individuals and 10 patients with AA (aplastic anemia). In the AA bone marrow group, cell proliferation activity and CFU-mix colonies of D and E fractions in experimental group 2 were significantly higher than those in negative control group and experimental group. The expression of Bcl-2: The experimental group 2 D, E group was significantly higher than the negative control group and experimental group 1. Mutant p53 expression between the three groups no significant difference. Conclusion Compared with the velvet antler of two different processes, the cold process velvet has a stronger biological effect, so the cold process is recommended in the processing of velvet antler.
