目的探讨新型H1N1流感病毒从2009年在苏州出现至2011年基本消失的整个过程中,苏州分离株的神经氨酸酶(Neuraminidase,NA)的分子特征及其基因变异和发展规律。方法提取苏州分离株的RNA,以WHO推荐的引物扩增NA基因并测序。以WHO推荐的季节性H1N1和新型H1N1疫苗株为参照,采用生物信息学软件对苏州毒株的NA进行详细的分子特征分析,并与全球及全国的代表性毒株进行序列进化分析。结果与疫苗株相比,5株苏州株的核苷酸序列同源性均值为99.64%,氨基酸序列同源性均值为99.36%。5株苏州株中都没有发现抗原位点的改变,但4株在耐药性相关位点附近出现N248D突变,还有1株2011年株增加了1个新的潜在糖基化位点。结论首次详细分析了苏州地区2009年以来H1N1型流感病毒NA的分子特征,表明苏州毒株出现了一个新的潜在糖基化位点,但抗原性和药物敏感性均保持稳定。
Objective To investigate the molecular characteristics of neuraminidase (NA) and its genetic variation and development in Suzhou isolates from the year 2009 when the new H1N1 influenza virus emerged from Suzhou to almost disappear in 2011. Methods The RNA of Suzhou isolate was extracted. The NA gene was amplified with the primers recommended by WHO and sequenced. Using the seasonal H1N1 and H1N1 vaccine strains recommended by WHO as reference, the bioinformatics software was used to analyze the molecular characteristics of NA in Suzhou strain in detail and sequence evolution analysis was carried out with the representative strains of the whole country and the whole country. Results Compared with the vaccine strain, the five strains of Suzhou had 99.64% nucleotide homology and 99.36% amino acid sequence homology. No mutation of the antigenic site was found in the 5 strains of Suzhou, but 4 strains showed N248D mutation in the vicinity of resistance-related sites and 1 strain added 1 new potential glycosylation site. Conclusions The molecular characteristics of NA of H1N1 influenza virus in Suzhou area since 2009 are analyzed in detail for the first time. It indicates that there is a new potential glycosylation site in Suzhou strain, but both antigenicity and drug sensitivity remain stable.
