为探讨旋毛虫p43、p53基因的核酸疫苗对小鼠的免疫保护效果与免疫保护机制,将p43、p53基因分别克隆到真核表达载体pcDNA3.1(+)上,分3次肌肉注射免疫3组昆明小鼠。于三免后2周经口感染旋毛虫肌幼虫(100蚴/只),在攻虫后第7天和第35天剖杀小鼠,计算成虫减虫率、肌幼虫减虫率和繁殖力指数,评估其免疫效果。在各时间点上取小鼠血清用于检测抗体水平和脂肪酸结合蛋白(H-FABP)。结果显示,试验各组成虫减虫率与对照组相比差异显著(P<0.05),LPG及RCI减虫率与对照组相比均差异极显著(P<0.01);IgG抗体水平提高,与PBS组差异显著(P<0.05);各组H-FABP检测结果均低于70pg/mL。结果表明旋毛虫p43与p53基因的核酸疫苗具有较好的抗旋毛虫的免疫保护效果,且两核酸疫苗对小鼠心肌无损伤。
In order to explore the immunoprotective effect and immunoprotection mechanism of p43 and p53 gene DNA vaccine in mice, the p43 and p53 genes were cloned into the eukaryotic expression vector pcDNA3.1 (+) respectively and immunized 3 times intramuscularly Kunming mice. Trichinella spiralis larvae (100 larvae / bird) were orally infected 2 weeks after the three immunizations and mice were sacrificed on the 7th and 35th days after the helminth to calculate the worm reduction rate, worm larva reduction rate and fecundity Index, to assess the immune effect. The H-FABP test results were lower than 70 pg / mL in each group. The results showed that Trichinella spiralis p43 and p53 gene nucleic acid vaccine has good immunological protection against Trichinella spiralis, and the two nucleic acid vaccine did not damage the myocardium of mice.
