很早以来,在临床及病理工作中就有很多人观察到不少由肝炎、肝硬化最后发展成肝癌的病例,由于缺乏病原学依据,未能深入研究其间的联系。1965年Blumberg发现了乙型肝炎表面抗原(HBsAg)后,国内外许多学者利用血清HBsAg测定进行了流行病学方面的各种调查。从世界各地区获得的结果,均一致肯定了乙型肝炎病毒(HBV)与原发性肝癌的密切关系。随着免疫电镜、免疫荧光,免疫酶标等技术用于肝细胞内的乙型肝炎抗原测定,在敏感性和特异性上都有很大提高。但由于设备条件要求较高,操作步骤也很复杂,难于推广采用,更不能常规检测。 1974年日本志方俊夫(Shikata)用改良地衣红(Orcoin)染色法在常规甲醛固定的肝组织石腊切片中,发现肝细胞胞浆内HBsAg聚集物与地衣红结合显示出特殊颜色,以此可判定肝细胞内HBV感染。经其本人和后来许多学者用免疫荧光法,过氧化酶等作对照,
