研究眼镜蛇蛇毒组分C诱导白血病细胞凋亡的作用及机制。方法:应用光学显微镜、透射电镜、脱氧核糖核酸( DNA)电泳、流式细胞仪、逆转录聚合酶链反应(RT-PCR)等方法,观察白血病细胞经过眼镜蛇蛇毒组分C处理后,形态学、生物化学及BcI-2/Bax基因表达水平的变化。结果:眼镜蛇蛇毒组分C能诱导人粒细胞白血病细胞系(HL60)发生凋亡形态学和生物化学变化;流式细胞议分析发现眼镜蛇蛇毒组分C能使一定比例的人粒单细胞白血病细胞系(J6-1)、人红白血病细胞系(K562)、HL60。细胞及取自患者骨髓的白血病细胞发生凋亡,而且凋亡率随蛇毒浓度的增高而增加;RT-PCR方法检测发现眼镜蛇蛇毒组分C能使HL60细胞Bcl-2基因的表达下调,而Bax变化不明显。结论:眼镜蛇蛇毒组分C能诱导白血病细胞发生凋亡,此作用与Bcl-2基因的表达下调有关。
To study the effect and mechanism of cobra venom component C on apoptosis of leukemia cells. METHODS: The morphology of leukemia cells treated with cobra venom component C was observed by light microscopy, transmission electron microscopy, DNA electrophoresis, flow cytometry, and reverse transcription polymerase chain reaction (RT-PCR). , biochemistry and changes in BcI-2/Bax gene expression levels. RESULTS: Cobra venom component C could induce morphological and biochemical changes of apoptosis in human HL60 cell line. Flow cytometry analysis showed that component C of cobra venom can cause a certain proportion of human granulocyte-monocyte leukemia cells. Department (J6-1), human erythroleukemia cell line (K562), HL60. Cells and leukemic cells taken from the patient’s bone marrow undergo apoptosis, and the apoptotic rate increases with the increase of venom concentration. The RT-PCR method found that Cobra venom component C can down-regulate the expression of Bcl-2 gene in HL60 cells, while Bax The change is not obvious. Conclusion: Cobra venom component C can induce apoptosis of leukemic cells, and this effect is related to the down-regulation of Bcl-2 gene expression.
