目的 研究p16基因改变与脑胶质瘤恶性程度分级及肿瘤细胞增殖活性之间的关系。方法 检测 41例不同级别脑胶质瘤标本和 7例正常脑组织p16基因改变和PCNA蛋白表达情况。结果 实验组p16基因缺失和突变 2 2例 (5 3.7% ) ,mRNA和蛋白表达缺失分别为 2 3例 (5 6 .10 % )、2 7例 (6 5 .85 % ) ,三者改变非一对一关系。p16基因改变和PCNA指数随脑胶质瘤恶性程度级别增高呈上升趋势 (P <0 .0 5 )。p16基因改变与PCNA指数具有明显相关性 (P <0 .0 1)。结论 p16基因改变所导致的细胞周期调控失常及细胞增殖活性升高 ,是影响脑胶质瘤生物学行为的重要因素。
Objective To study the relationship between the alteration of p16 gene and the grade of malignant glioma and the proliferation of tumor cells. Methods The changes of p16 gene and the expression of PCNA protein in 41 cases of gliomas of different grades and 7 cases of normal brain tissue were detected. Results The deletion and mutation of p16 gene in the experimental group were 22 (53.7%), 23 (56.10%) and 27 (65.85% One to one relationship. The changes of p16 gene and PCNA index increased with the malignant degree of glioma (P <0.05). There was a significant correlation between p16 gene expression and PCNA index (P <0.01). Conclusion The abnormal cell cycle regulation and cell proliferative activity caused by the alteration of p16 gene are important factors affecting the biological behavior of glioma.
