目的 研究人脑胶质瘤细胞系对不同强度X线的敏感性。方法 以BT32 5细胞系为实验对象 ,设置了阴性对照、5Gy、10Gy、15Gy、2 0Gy和 30Gy组 ,应用Varian直线加速器行单次大剂量照射。采用免疫组化法观察p5 3、BcL 2和PCNA基因的表达情况 ,采用光镜和电镜观察细胞凋亡的形态学改变。结果 2 0Gy组 48h时 ,细胞凋亡发生率最高 ,此时p5 3基因表达最高 ,BcL 2基因表达明显减弱 ,但PCNA 12h内阳性率最高 ,随着时间延长 ,其表达率明显下降 ,尤以 48h为著。结论 p5 3、BcL 2和PCNA是与放射效应密切相关的基因
Objective To study the sensitivity of human glioma cell lines to different intensities of X-rays. Methods BT32 5 cell lines were used as experimental subjects, and negative control, 5Gy, 10Gy, 15Gy, 20Gy and 30Gy groups were set up and single bolus irradiation was performed with Varian linear accelerator. Immunohistochemistry was used to observe the expression of p53, Bcl-2 and PCNA genes. Morphological changes of apoptosis were observed under light microscope and electron microscope. Results At 48 hours after 20 Gy treatment, the highest incidence of apoptosis was found. At this time, the expression of p5 3 gene was the highest while the expression of Bcl-2 gene was significantly decreased. However, the positive rate of PCNA within 12 hours was the highest, with a significant decrease over time 48h for. Conclusion p5 3, BcL 2 and PCNA are closely related to the radiation effects of genes
