目的:探讨川芎嗪对过氧化氢引起PC12细胞损伤的保护作用。方法:采用大鼠肾上腺嗜铬瘤克隆化细胞株PC12细胞,制备过氧化氢细胞损伤模型,通过CCK-8法检测细胞活性、流式细胞术测定线粒体膜电位和酶联免疫吸附测定(ELISA)检测炎症性细胞因子、氧化应激指标观察过氧化氢的毒性及川芎嗪的保护作用。结果:川芎嗪预处理后,PC12细胞的存活率显著提高,细胞线粒体膜电位提高,白介素1β(IL-1β)分泌水平显著降低,超氧化物歧化酶(SOD)上调,乳酸脱氢酶(LDH)和丙二醛(MDA)下调,谷胱甘肽(GSH)升高。结论:川芎嗪对过氧化氢引起的PC12细胞损伤有显著的保护作用,其保护作用可能与川芎嗪抑制细胞凋亡、调节氧化应激反应相关。
Objective: To investigate the protective effect of ligustrazine on hydrogen peroxide-induced PC12 cell injury. Methods: The cell model of hydrogen peroxide injury was established by clonogenic cell line of rat adrenal pheochromocytoma cell line PC12. The cell viability was measured by CCK-8 assay. Mitochondrial membrane potential and enzyme linked immunosorbent assay (ELISA) were determined by flow cytometry. To detect inflammatory cytokines, oxidative stress indicators to observe the toxicity of hydrogen peroxide and tetramethylpyrazine protective effect. Results: After pretreatment with ligustrazine, the survival rate of PC12 cells was significantly increased, the membrane potential of mitochondria was increased, the level of IL-1β was significantly decreased, the level of superoxide dismutase (SOD) was up-regulated, and lactate dehydrogenase ) And malondialdehyde (MDA), glutathione (GSH) increased. CONCLUSION: Tetramethylpyrazine has a significant protective effect on hydrogen peroxide-induced injury of PC12 cells, and its protective effect may be related to the inhibition of apoptosis and the regulation of oxidative stress by ligustrazine.
