目的了解北京台湖地区淡色库蚊对常用杀虫剂的抗药情况和当地淡色库蚊kdr基因多态性,为该地区淡色库蚊防治提供科学指导。方法采用幼虫浸渍法对氯菊酯、残杀威和毒死蜱进行抗药性生物检测,以等位基因特异性PCR(AS-PCR)为检测方法确定当地淡色库蚊kdr基因型的频率。结果当地淡色库蚊对氯菊酯、残杀威和毒死蜱的抗性系数分别为15.9、4.2和3.6;当地淡色库蚊中kdr基因存在一种突变(L1014F型),突变基因占基因总数的25.7%。结论北京台湖地区的淡色库蚊已对氯菊酯产生较强抗性,建议在以后的防治工作中通过杀虫剂的复配和交替用药达到蚊虫防治目的。
Objective To understand the resistance of Culex pipiens pallens to common insecticides and the kdr gene polymorphism of Culex pipiens pallens in the Taihu Lake in Beijing, and to provide scientific guidance for the prevention and control of Culex. Methods Permethrin, propoxur and chlorpyrifos were tested for drug resistance by larvae dipping method. The kdr genotype frequency of Culex pipiens pallens was determined by allele-specific PCR (AS-PCR) method. Results The resistance coefficients of Culex pipiens pallens to chlorpromazine, propoxur and chlorpyrifos were 15.9, 4.2 and 3.6, respectively. There was a mutation (L1014F) in kdr gene in Culex pipiens pallens, which accounted for 25.7% . Conclusion Culex pipiens pallens in Taiwan Lake area of Beijing has a strong resistance to permethrin. It is suggested that mosquito control purposes should be achieved through the compounding and alternate application of insecticides in the future prevention and treatment.
